The bus
Use the Nueva Central Camionera, Guadalajara''s long-distance terminal, on the Tlaquepaque side of the city — not the old central camionera, which handles regional routes. It has several modules; confirm which one your carrier uses before you set off, because they are a walk apart.
Primera Plus and ETN are the first-class names on this corridor, with departures through the day and assigned seats. The ride is about four and a half hours, usually with a stop in León or Irapuato. Buy on the carrier''s app when you can; weekend and holiday services do sell out.
Morning departures are worth the early alarm — you arrive with the afternoon intact instead of dragging luggage through the callejones after dark.
Driving
Leave Guadalajara east on the 15D toward Zapotlanejo, then continue on the 45D through Irapuato and Silao before the Guanajuato spur. Around 300 km, about four hours, all cuota.
Take the toll road the whole way and keep the tank above half — service stations are spaced out across the Bajío. Drive in daylight; this is a busy freight corridor and the libre alternatives are slow and poorly lit at night.

Arriving in Guanajuato
The bus terminal is about 6 km west of the historic centre. Taxis wait outside — agree the fare or buy a ticket at the booth — and the ride takes 15 to 20 minutes. The local city bus marked Centro is far cheaper and fine with a backpack.
Give the driver a landmark rather than an address: Jardín Unión, Plaza de la Paz, Teatro Juárez, Mercado Hidalgo. Many hotels are up alleys no car can enter.
